Marsden got his first job in 1993 on a series of episodes of The Nanny as Eddie, Margaret Sheffield's boyfriend.
In 2007, Marsden played Corny Collins in the film adaptation of the Broadway musical based on the 1988 John Waters movie Hairspray alongside Michelle Pfeiffer, Zac Efron, Nikki Blonsky, Amanda Bynes, Queen Latifah and John Travolta. There he also made his musical debut by singing two of the film's songs, "The Nicest Kids In Town" and "(It's) Hairspray." Both "Hairspray" and the film's soundtrack were critically acclaimed. The soundtrack has sold over 1,200,000 copies to date in the United States, and has been certified Platinum by the RIAA. His next role was in the Disney hybrid animated/live action film Enchanted, playing Prince Edward with co-stars Amy Adams, Susan Sarandon, Idina Menzel, Timothy Spall and Patrick Dempsey. He sang one song as a duet with Amy Adams at the beginning of the film and a duet with Idina Menzel that was cut from the film. '

The song "Glow" by James Marsden seems to be a tribute to the power of love and happiness that can make a person feel like they're glowing. The first verse speaks of the contrast between the confined feeling of being stuck indoors, not doing anything, to the adventurous feeling of being out there in the world, kicking faster than a modern disaster, and flying higher than any pilot, shooting Stardust over open sky. The singer of the song likens this to the feeling of being in love - when you're with the person you love, everything else fades away, and you feel like you're soaring.
The second verse uses similar imagery, with the singer feeling weightless in zero gravity, having never loved anything in that space before. Out there, though, their love interest is making even more incredible feats of daring, racing backwards like a racecar driver, holding up the runaway train, and tearing petals from their new violet, making wishes everyone already knows. The whole song closes with the singer confessing that they'll glow for their love interest if their love interest will do the same for them - a mutually supportive relationship fueled by the power of love.
