Nasceu em uma família rica e tradicional, e casou aos 18 anos com um herdeiro da milionária família paulista Matarazzo, 20 anos mais velho que ela. Desde antes do casamento já compunha e tocava piano, e mesmo depois continuou cantando suas músicas em festinhas íntimas de amigos.
Quando um produtor musical a ouviu, quis contratá-la imediatamente para gravar um disco, mas Maysa, que estava grávida, pediu que esperasse o nascimento do filho. Afinal saiu "Convite para Ouvir Maysa", em quatro volumes, entre 1956 e 59, com os sucessos "Ouça", "Adeus" e "Meu Mundo Caiu".
Tornou-se uma estrela, para desgosto da família do marido, e separou-se pouco depois, ficando abalada e deprimida com o fato. Suas músicas, já tradicionalmente de "fossa", tornaram-se ainda mais melancólicas
Maysa compôs 26 músicas, numa época em que era pouco comum a existência de mulheres compositoras. Com uma vida sempre agitada por casos amorosos e problemas com bebida, Maysa gravou alguns dos discos mais importantes da bossa nova e da música romântica brasileira.
Fez teatro e televisão, tendo atuado na telenovela O Cafona, com Francisco Cuoco, e na telenovela Bravo, com Carlos Alberto.
Em janeiro de 1977, a cantora morreu num trágico acidente de automóvel na Ponte Rio-Niterói onde o carro que ela dirigia colidiu com a barra de concreto que separa as pistas. É bastante provável que o acidente tenha sido, na realidade, um suicídio.
