Foi locutor de rádio em Cachoeiro de Itapemirim (ES), onde nasceu. Na década de 60 mudou-se para o Rio de Janeiro, onde continuou atuando como radialista. Em 1970 lançou-se como cantor, dividindo com Raul Seixas, Miriam Batucada e Eddy Star o disco "Sociedade da Grã-Ordem Kavernista Apresenta: Sessão das Dez". Lançou um disco em 1972 com a música "Eu Quero É Botar Meu Bloco na Rua", com a qual classificou-se para o VII Festival Internacional na Canção. Este foi um monumental sucesso de vendagem que ele não conseguiria repetir. Gravou em 1976 o LP "Tem Que Acontecer", pela Continental e em 1978 "Sinceramente", este por uma gravadora independente. Em 1998 o compositor Sergio Natureza produziu o CD tributo "Balaio do Sampaio", uma homenagem póstuma em que vários intérpretes executam as suas composições. Em 2000 foi lançada biografia de autoria de Rodrigo Moreira, pela editora Muiraquitã.
O músico maranhense Zeca Baleiro lançou o disco "Cruel", com as composições inéditas que seriam lançadas no quarto disco de Sérgio Sampaio e que haviam ficado, também num 'baú', nas famosas 'fitinhas'.
No final de 2009 o músico Juliano Gauche (também com a banda Solana), juntamente com o Duo Zebedeu (Fábio do Carmo e Júlio Santos) lançaram o CD HOJE NÃO, com canções de composição de Sérgio Sampaio e produção de João Moraes. Juliano e o Duo saem em turnê em março de 2010.
