Foi criado em Nova Europa, cidade do interior de São Paulo, onde morou até 1954; depois mudou-se com a família para São Carlos, onde pode começar sua carreira musical, pois a cidade possuia a melhor vida noturna de toda a região.
Sua carreira musical começou quando foi crooner no meio dos anos 50 no interior de São Paulo, na cidade de São Carlos, lá chegando em 1954 e participando da noite sãocarlense que era intensa na época, também com participações na Rádio São Carlos como calouro e com apresentações, vivendo intensamente em São Carlos, até o fim da década. No início da década seguinte foi tentar o sucesso na capital do Estado, e obteve-o participando de programas de calouros na televisão.
Elis Regina e Jair Rodrigues fizeram muito sucesso com sua parceria no programa O Fino da Bossa, programa da TV Record, em 1965.
Em 1966, Jair participou do festival daquele ano com a música Disparada, de Geraldo Vandré e Théo de Barros, desta vez em conjunto com o Quarteto Novo. Conhecido por cantar sambas, Jair surpreendeu o público com uma linda interpretação da canção. Disparada e Banda, de Chico Buarque e interpretada por Nara Leão, eram favoritas. O festival acabou empatado. A partir daquele momento, sua carreira decolou e seu talento assegurou décadas de sucesso ao cantor. Jair lançou um álbum por ano e interpretou sucessos como O Menino da Porteira, Boi da Cara Preta e Majestade o Sabiá. Realizou turnês pela Europa, Estados Unidos e Japão. Em 1971, gravou o samba-enredo Festa para um Rei Negro, da Acadêmicos do Salgueiro, do Rio de Janeiro.
Nas décadas seguintes, sua produção diminuiu de volume; entretanto, Jair Rodrigues continua conhecido por sua grande energia e sua alegria contagiante.
Faleceu em 8 de maio de 2014, aos 75 anos.
